    Experimental study of hybrid strengthening technique using carbon fiber laminates and steel plates for reinforced concrete slabs

    External bonding (EB) technique has been used widely to strengthen reinforced concrete (RC) structures in civil engineering over several decades. Steel and fiber reinforced polymers (FRP) are the two most common materials used for strengthening RC structures. Although each material has its advantages for EB application, they also have their own weaknesses. Steel plates are heavy and poor in corrosion resistance, and the thickness of steel plates is also limited due to lack of shape flexibility for strengthening RC structures. The EB-FRP technique is restricted for broader applications because the bond interface is the weakest link in such retrofitted or strengthened concrete members, resulting in sudden and brittle debonding failure, and the strength of FRP has not been fully utilized. To overcome these weaknesses, an innovative strengthening method combining FRP and steel materials was explored in this research. Carbon fiber laminates (CFL) and thin steel plates (SP) were combined in different ways to determine the most effective hybrid strengthening configuration. A total of nineteen RC slabs strengthened by different configurations were tested under four-point bending. Failure modes, load versus deflection curves, strains of internal reinforcement bars, CFL and SP, and stiffness of the strengthened specimens were analyzed. The test results proved that the CFL-SP hybrid strengthening technique was effective. The average maximum increases in flexural capacity and stiffness were 204.2% and 91% over the control slab, respectively. In addition, the cracking and steel yielding loads of slabs strengthened with CFL-SP composite material were higher than those of slabs strengthened by FRP or steel plate only. By combining with steel plate, FRP material was used more efficiently and the stiffness of the strengthened RC slabs was improved. The overlap-type hybrid configuration of bonding CFL as inner plates while steel as outer plates was proved as the most effective hybrid strengthening method within all configurations tested in this research

    Investigating the Role of FlhF Identifies Novel Interactions With Genes Involved in Flagellar Synthesis in Campylobacter jejuni.

    FlhF is a key protein required for complete flagellar synthesis, and its deletion results in the complete absence of a flagella and thus motility in Campylobacter jejuni. However, the specific mechanism still remains unknown. In this study, RNA-Seq, EMSAs, ChIP-qPCR and β-Galactosidase assays were performed to elucidate the novel interactions between FlhF and genes involved in flagellar synthesis. Results showed that FlhF has an overall influence on the transcription of flagellar genes with an flhF mutant displaying down-regulation of most flagellar related genes. FlhF can directly bind to the flgI promoter to regulate its expression, which has significant expression change in an flhF mutant. The possible binding site of FlhF to the flgI promoter was explored by continuously narrowing the flgI promoter region and performing further point mutations. Meanwhile, FlhF can directly bind to the promoters of rpoD, flgS, and fliA encoding early flagellin regulators, thereby directly or indirectly regulating the synthesis of class I, II, and III flagellar genes, respectively. Collectively, this study demonstrates that FlhF may directly regulate the transcription of flagellar genes by binding to their promoters as a transcriptional regulator, which will be helpful in understanding the mechanism of FlhF in flagellar biosynthetic and bacterial flagellation in general

    Evolution of the class C GPCR Venus flytrap modules involved positive selected functional divergence

    <p>Abstract</p> <p>Background</p> <p>Class C G protein-coupled receptors (GPCRs) represent a distinct group of the GPCR family, which structurally possess a characteristically distinct extracellular domain inclusive of the Venus flytrap module (VFTM). The VFTMs of the class C GPCRs is responsible for ligand recognition and binding, and share sequence similarity with bacterial periplasmic amino acid binding proteins (PBPs). An extensive phylogenetic investigation of the VFTMs was conducted by analyzing for functional divergence and testing for positive selection for five typical groups of the class C GPCRs. The altered selective constraints were determined to identify the sites that had undergone functional divergence via positive selection. In order to structurally demonstrate the pattern changes during the evolutionary process, three-dimensional (3D) structures of the GPCR VFTMs were modelled and reconstructed from ancestral VFTMs.</p> <p>Results</p> <p>Our results show that the altered selective constraints in the VFTMs of class C GPCRs are statistically significant. This implies that functional divergence played a key role in characterizing the functions of the VFTMs after gene duplication events. Meanwhile, positive selection is involved in the evolutionary process and drove the functional divergence of the VFTMs. Our results also reveal that three continuous duplication events occurred in order to shape the evolutionary topology of class C GPCRs. The five groups of the class C GPCRs have essentially different sites involved in functional divergence, which would have shaped the specific structures and functions of the VFTMs.</p> <p>Conclusion</p> <p>Taken together, our results show that functional divergence involved positive selection and is partially responsible for the evolutionary patterns of the class C GPCR VFTMs. The sites involved in functional divergence will provide more clues and candidates for further research on structural-function relationships of these modules as well as shedding light on the activation mechanism of the class C GPCRs.</p

    TiEV: The Tongji Intelligent Electric Vehicle in the Intelligent Vehicle Future Challenge of China

    TiEV is an autonomous driving platform implemented by Tongji University of China. The vehicle is drive-by-wire and is fully powered by electricity. We devised the software system of TiEV from scratch, which is capable of driving the vehicle autonomously in urban paths as well as on fast express roads. We describe our whole system, especially novel modules of probabilistic perception fusion, incremental mapping, the 1st and the 2nd planning and the overall safety concern. TiEV finished 2016 and 2017 Intelligent Vehicle Future Challenge of China held at Changshu. We show our experiences on the development of autonomous vehicles and future trends

    Quantitative Comparisons of Outer-Rotor Permanent Magnet Machines of Different Structures/Phases for In-Wheel Electrical Vehicle Application

    As one of the key components, low-speed direct-drive in-wheel machines with high compact volume and high torque density are important for the traction system of electric vehicles (EVs). This paper introduces four different types of outer-rotor permanent magnet motors for EVs, including one five-phase SPM machine, one three-phase IPM machine with V-shaped PMs, one seven-phase axial flux machine (AFM) of sandwich structure and finally one hybrid flux (radial and axial) machine with a third rotor with V-shaped PMs added to the AFM. Firstly, the design criteria and basic operation principle are compared and discussed. Then, the key properties are analyzed using the Finite Element Method (FEM). The electromagnetic properties of the four fractional slot tooth concentrated winding in-wheel motors with similar dimensions are quantitatively compared, including air-gap flux density, electromotive force, field weakening capability, torque density, losses, and fault tolerant capability.     The Novel Protein Cj0371 Inhibits Chemotaxis of Campylobacter jejuni

    cj0371 is a novel gene that is associated with Campylobacter jejuni virulence, and an isogenic mutant of cj0371 showed hyper chemotaxis and motility. Chemotactic motility is an important virulence factor and is involved in C. jejuni pathogenesis. Campylobacter sp. has specific variations of the common chemotaxis components, including histidine autokinase CheA, coupling scaffold protein CheV, chemotaxis response regulator protein CheY and several chemoreceptor proteins. In this study, we used immunoprecipitation combined with LC-MS/MS analyses to screen six chemotaxis pathway proteins that potentially interact with the putative protein Cj0371. qRT-PCR was used to quantitatively analyze the expression of these chemotaxis genes and basic flagella genes. The results showed that the expression of cheV, cj1110c, and cj0262c was significantly up-regulated, and four flagella genes also had up-regulated expression in the cj0371 mutant. GST pull-down analyses found that Cj0371 interacted with the receiver domain of the CheV protein. Enzyme-coupled spectrophotometric assays showed that the ATPase activity of CheA was higher when Cj0371 was not present in the chemotaxis reaction medium. Therefore, we concludes that cj0371 has a negative influence on C. jejuni chemotaxis, which may occur by adjusting the receiver domain of CheV to influence chemotaxis. This paper provides a new component in the chemotaxis pathway of C. jejuni for the first time and highlight the complexity of this remarkable pathway
